<title>Ringkasan</title>
<p>Pinang (Areca catechu L) secara tradisional dimanfaatkan untuk mengobati luka dan pendarahan, infeksi saluran kemih, sakit kaki dan kecacingan. Studi penelitian modern, pinang memiliki efek farmakologis sebagai antijamur. Penelitian ini bertujuan untuk mengetahui kemampuan ekstrak etanol biji pinang terhadap pertumbuhan jamur Candida albicans. Penelitian ini merupakan penelitian eksperimen. Langkah-langkah yang dilakukan dalam penelitian ini, mula-mula biji pinang yang telah dikupas dijemur selama 7-10 hari. Setelah kering, dihaluskan menjadi serbuk. Selanjutnya proses ekstraksi fitokimia menggunakan metode maserasi menggunakan etanol. Perolehan ekstrak dilakukan menggunakan rotary vacuum evaporator. Ekstrak yang diperoleh dibuat taraf konsentrasi 20, 40, 60 dan 80 g/ml. Masing-masing taraf konsentrasi dilakukan uji daya hambat anti jamur menggunakan metode <italic>Kirby Bauer</italic>. Zona bening yang terbentuk diukur menggunakan jangka sorong. Data dianalisis menggunakan statistik non Parametrik <italic>Kruskal Wallis</italic> dan uji lanjut <italic>Mann-Whitney U</italic>. Secara deskriptif daya hambat ekstrak etanol biji pinang memiliki kemampuan daya hambat terhadap jamur C. albicans. Analisis statistik semua taraf konsentrasi memberikan pengaruh yang nyata dalam menghambat pertumbuhan jamur. Uji lanjut menunjukkan konsentrasi 20 g/ml dan 80  g/ml serta 60 g/ml dan 80 g/ml memiliki nilai signifikasinya &lt;0,05, yang berarti ada perbedaan pengaruh dalam menghambat pertumbuhan jamur C. albicans. Kesimpulan penelitian ini adalah ekstrak etanol biji pinang dapat menghambat pertumbuhan jamur C. albicans.</p>

<title>Abstract</title>
<p>Areca nut (Areca catechu L) is traditionally used to treat wounds and bleeding, urinary tract infections, leg pain, and worms. Modern research studies, the areca nut has a pharmacological effect as an antifungal. This study aims to determine the ability of the ethanol extract of areca nut to the growth of the fungus Candida albicans. This research is experimental research. The steps are taken in this study, firstly, the peeled areca nut seeds were dried in the sun for 7-10 days. After drying, crushed into powder. Furthermore, the phytochemical extraction process uses the maceration method using ethanol. Extracts were obtained using a rotary vacuum evaporator. The extracts obtained were made in series with concentrations of 20, 40, 60, and 80 g/ml. Each concentration series was tested for antifungal inhibition using the Kirby Bauer method. The clear zone formed was measured using a calliper. The data were analyzed using non-parametric Kruskal Wallis statistics and Mann-Whitney U test. The results of the inhibitory test of areca nut ethanol extract showed descriptively that the ethanol extract had the inhibitory ability against the fungus C. albicans. Statistical analysis of all concentration levels gave a significant effect in inhibiting fungal growth. Further tests showed that the concentrations of 20 g/ml and 80 g/ml as well as 60 g/ml and 80 g/ml had a significance value of &lt;0.05, which means that there were different effects in inhibiting the growth of the fungus C. albicans. This study concludes that the ethanol extract of areca nut seeds can inhibit the growth of the fungus C. albicans.</p>

<bold>PENDAHULUAN</bold>
</title>
<p>Pinang (<italic>Areca catechu</italic>
<italic>L.</italic>) merupakan salah satu jenis tanaman palem, yang terdistribusi luas di wilayah Afrika, daerah tropis Asia, termasuk Indonesia. Tanaman ini secara tradisional oleh masyarakat dimanfaatkan untuk mengobati luka dan pendarahan, infeksi saluran kemih, sakit kaki, menghilangkan cairan dalam rongga perut dan kecacingan (<xref ref-type="bibr" rid="redalyc_5043270002_ref9">Essien et al., 2017</xref>; <xref ref-type="bibr" rid="redalyc_5043270002_ref12">Jaiswal et al., 2011</xref>; <xref ref-type="bibr" rid="redalyc_5043270002_ref20">Peng et al., 2015</xref>). Di Papua, pinang dijadikan sebagai cemilan bahkan telah menjadi kebiasaan yang secara turun temurun diwariskan. Kebiasaan mengunyah biji pinang dapat bertindak sebagai stimulan, memperkuat gigi dan gusi serta dapat mengurangi nafsu makan.</p>
<p>Studi penelitian modern tentang pinang, membuka cakrawala baru tentang potensi pinang sebagai sumber alternatif pengobatan berbagai penyakit. Puluhan kandungan senyawa kimia yang telah teridentifikasi diduga kuat memiliki banyak aktivitas farmakologis, termasuk anti diabetes, mengurangi kadar kolesterol (<xref ref-type="bibr" rid="redalyc_5043270002_ref4">Anthikat &amp; Michael, 2009</xref>), anti parasit, anti inflamasi, anti depresi, memiliki efek analgesik, anti bakteri, antifungi (<xref ref-type="bibr" rid="redalyc_5043270002_ref20">Peng et al., 2015</xref>), anti-HIV, antiaging, anti alergi (<xref ref-type="bibr" rid="redalyc_5043270002_ref2">Amudhan et al., 2012</xref>), antioksidan, anti kecacingan (<xref ref-type="bibr" rid="redalyc_5043270002_ref19">Patil et al., 2009</xref>) dan anti malaria (<xref ref-type="bibr" rid="redalyc_5043270002_ref3">Ansari et al., 2021</xref>).</p>
<p>Jamur Candida (<italic>Candida albicans</italic>) merupakan patogen pada manusia. <italic>Candida albicans</italic> mendiami berbagai permukaan tubuh seperti rongga mulut, saluran pencernaan, vagina, dan kulit, tanpa menimbulkan masalah pada individu yang sehat, namun dalam kondisi tertentu dapat menyebabkan infeksi superfisial dan infeksi sistemik (<xref ref-type="bibr" rid="redalyc_5043270002_ref8">Calderone &amp; Clancy, 2011</xref>; <xref ref-type="bibr" rid="redalyc_5043270002_ref26">Teodoro et al., 2015A</xref>). Salah satu sifat infeksius yang dimiliki jamur Candida adalah memampuan beradaptasi dengan perubahan lingkungan dengan mengubah sifat fenotip (<italic>switch phenotype</italic>). Disamping itu sekresi enzim lipase, esterase, hemolisin, proteinase aspartil, menambah daya invasi Candida dalam jaringan tubuh (<xref ref-type="bibr" rid="redalyc_5043270002_ref11">Jabra-Rizk et al., 2016</xref>).</p>
<p>Salah satu problematika dalam penanganan dan pengendalian penyakit infeksi adalah resistensi terhadap obat (<italic>antimicrobial drug resistance)</italic>, termasuk kasus resistensi pada jamur <italic>C. albicans</italic> kemampuan membentuk biofilm yang memungkinkanya dapat menghindari sistem pertahanan inang. Permasalahan resistensi ini menambah beban dalam upaya pengendalian penyakit infeksi (<xref ref-type="bibr" rid="redalyc_5043270002_ref28">Vadhana et al., 2015</xref>). Diperlukan sebuah pendekatan holistik untuk mengurai permasalahan ini, salah satu alternatif yang ditempuh adalah eksplorasi senyawa-senyawa yang terkandung dalam tanaman. Hal ini didukung dengan sebuah estimasi bahwa sekitar 80% penduduk di negara berkembang masih memanfaatkan obat tradisional untuk kesehatan (<xref ref-type="bibr" rid="redalyc_5043270002_ref1">Al-Bayati, 2016</xref>), juga beberapa jenis tanaman secara organoleptik mudah diterima oleh masyarakat.</p>
<p>Beberapa tanaman dilaporkan dapat menghambat pertumbuhan <italic>C. albicans.</italic> Tanaman yang dimaksud diantaranya <italic>Etlingera flexuosa</italic> (<xref ref-type="bibr" rid="redalyc_5043270002_ref21">Pitopang et al., 2020</xref>), daun kamboja putih atau <italic>Plumeria acuminata</italic> (<xref ref-type="bibr" rid="redalyc_5043270002_ref24">Sari et al., 2019</xref>), daun <italic>Curcuma longa</italic> (<xref ref-type="bibr" rid="redalyc_5043270002_ref23">Pulungan, 2017</xref>), dan rimpang binahong atau <italic>Anredera cordifolia</italic> (<xref ref-type="bibr" rid="redalyc_5043270002_ref15">Kurniawan, 2009</xref>). Kemampuan beberapa tanaman ini dipengaruhi oleh senyawa bioaktif yang terkandung didalamnya. Tumbuhan secara umum merupakan sumber yang kaya dengan bioaktif metabolit sekunder seperti tanin, terpenoid, saponin, alkaloid, flavonoid, dan senyawa lainnya. Senyawa-senyawa tersebeut dilaporkan secara in vitro memiliki sifat antijamur (<xref ref-type="bibr" rid="redalyc_5043270002_ref6">Arif et al., 2009</xref>).</p>
<p>Pinang dapat menjadi alternatif untuk menjawab permasalahan penanganan penyakit infeksi. Penelitian ragam senyawa kimia mengungkap fitokimia biji pinang bersifat aktif terhadap mikroorganisme. Penelitian yang dilakukan (<xref ref-type="bibr" rid="redalyc_5043270002_ref4">Anthikat &amp; Michael, 2009</xref>), ekstrak air biji pinang dapat membunuh bakteri gram positif, dan <italic>C. albicans</italic>. Investigasi lain melaporkan ekstrak biji pinang memiliki aktivitas penghambatan terhadap jamur Candida (<xref ref-type="bibr" rid="redalyc_5043270002_ref18">Pahadia et al., 2013</xref>). Senyawa fenol dan tanin pada ekstrak pinang memiliki aktivitas mikroba (<xref ref-type="bibr" rid="redalyc_5043270002_ref2">Amudhan et al., 2012</xref>; <xref ref-type="bibr" rid="redalyc_5043270002_ref31">Zhang et al., 2011</xref>). Penelitian ini bertujuan untuk mengetahui kemampuan bioaktivitas ekstrak etanol biji pinang terhadap jamur patogen terhadap manusia <italic>C. albicans.</italic>

<sec sec-type="materials|methods">
<title>
<bold>METODE</bold>
</title>
<sec>

<bold>Jenis Penelitian</bold>
</title>
<p>Penelitian ini merupakan penelitian eksperimen laboratorium.</p>
</sec>
<sec>
<title>
<bold>Lokasi dan Waktu Penelitian</bold>
</title>
<p>Penelitian ini dilaksanakan di Laboratorium Terpadu Poltekkes Kemenkes Jayapura pada bulan Mei-Juni 2021.</p>
</sec>
<sec>
<title>
<bold>Preparasi Bahan</bold>
</title>
<p>Preparasi bahan penelitian dimulai dengan proses pemisahan biji pinang dengan lapisan kulit luar, biji pinang dibelah menjadi beberapa bagian, selanjutnya dijemur di sinar matahari untuk mengurangi pertumbuhan jamur. Proses penjemuran dilakukan selama 7-10 hari, sampai tampak biji pinang kering mengeras dan berwarna merah kehitam-hitaman.</p>
<p>
<bold>Pembuatan Ekstrak</bold>
</p>
<p>Biji pinang yang telah dikeringkan ditumbuk dan diblender menjadi berbentuk serbuk. Serbuk biji pinang dimaserasi selama 72 jam menggunakan pelarut etanol 92% perbandingan 1:3.</p>
</sec>
<sec>
<title>
<bold>Maserasi, Evaporasi dan Pembuatan Larutan Sampel</bold>
</title>
<p>Ekstrak cair proses maserasi diperoleh menggunakan alat <italic>rotary vacuum evaporator</italic>, selanjutnya diuapkan dengan hot plate sampai didapatkan ekstrak yang kental. Larutan sampel ekstrak biji pinang dibuat empat tingkatan konsentrasi, yaitu 20, 40, 60, dan 80 g/ml, menggunakan aquades sebagai pelarut.</p>
</sec>
<sec>
<title>
<bold>Pengujian Daya Hambat</bold>
</title>
<p>Uji daya hambat ekstrak etanol biji pinang terhadap jamur <italic>C. albicans</italic> menggunakan metode <italic>Kirby Bauer</italic>. Mula-mula kultur yang telah diremajakan dicelupkan dalam tabung larutan isotonik, lalu dibandingkan kekeruhannya dengan larutan Mc Farlan 0,5. Biakan jamur <italic>C. albicans</italic> diapus sampai merata pada permukaan media SDA. Setelah apusan telah merata diseluruh permukaan media, kertas cakram yang dibasahi dengan 20 μl ekstrak pinang dan ketokenazol 2% sebagai kontrol positif diletakan diatas permukaan media. Selanjutnya semua perlakuan diinkubasi selama 24 jam pada suhu 37.C. Parameter yang diukur dalam pengujian antijamur adalah perubahan visual besarnya diameter zona bening yang terbentuk.</p>

<bold>Teknik Analisis Data</bold>
</p>
<p>Data penelitian diolah menggunakan uji <italic>Kruskal Wallis</italic> dan uji lanjut <italic>Mann-Whitney U</italic>. pada tingkat kepercayaan 95% (α = 0,05).</p>
</sec>
</sec>
<sec sec-type="results">
<title>
<bold>HASIL</bold>
</title>
<p>Hasil pengujian daya hambat aktivitas antijamur ekstrak etanol biji pinang tiap taraf konsentrasi.</p>

<p>Rerata diameter zona hambat tertinggi terdapat pada taraf konsentrasi 80 g/ml sebesar 11,33 mm, sedangkan rerata diameter zona hambat terkecil terdapat pada taraf perlakuan konsentrasi 20 dan 40 g/ml yaitu 9,00 mm. Hasil Uji statistik <italic>Kruskal Wallis one way analysis of variance by ranks</italic>, semua perlakuan memiliki nilai signifikansi 0,000 &lt;0,05, yang berarti semua taraf konsentrasi ekstrak etanol biji pinang berpengaruh nyata menghambat pertumbuhan jamur <italic>C. albicans</italic>. Untuk melihat perbedaan perlakuan masing-masing kadar konsentrasi pada C. <italic>albicans</italic>, dilakukan analisis lanjut menggunakan uji <italic>Mann-Whitney U</italic>.</p>

<p>Konsentrasi 20 dan 80 g/ml serta 60 dan 80 g/ml nilai signifikasinya &lt;0,05, yang berarti ada perbedaan pengaruh ekstrak etanol biji pinang terhadap jamur <italic>C. albicans</italic>.</p>

<title>
<bold>PEMBAHASAN</bold>
</title>
<p>Berdasarkan hasil penelitian ekstrak etanol pinang dapat menghambat proliferasi jamur <italic>C. albicans</italic>, hal ini terlihat dari adanya zona bening yang terbentuk (<xref ref-type="fig" rid="gf1">Gambar 1</xref>). Zona bening yang terbentuk menunjukkan kemampuan senyawa bioaktif metabolit sekunder biji pinang terhadap jamur <italic>C. albicans</italic>. Secara deskriptif konsentrasi 80 g/ml memiliki daya hambat paling tinggi (<xref ref-type="table" rid="gt1">Tabel 1</xref> &amp; <xref ref-type="table" rid="gt2">2</xref>) disebabkan kandungan kuantitas senyawa bioaktif. Penetrasi senyawa bioaktif terhadap mikroba salah satunya ditentukan jumlah senyawa aktif, umumnya semakin besar senyawa bioaktif yang digunakan maka semakin besar pula kemampuan daya hambatnya. Kemampuan ekstrak etanol biji pinang dalam menghambat pertumbuhan jamur <italic>C.albicans</italic> dipengaruhi oleh senyawa metabolit sekunder yang terkandung dalam biji pinang. Kurang lebih terdapat 50 senyawa telah teridentifikasi, yang diantaranya diduga memiliki kemampuan antimikroba termasuk anti jamur. Diantara senyawa-senyawa tersebut adalah flavonoid, tanin, terpenoid, alkaloid dan beberapa asam lemak (<xref ref-type="bibr" rid="redalyc_5043270002_ref2">Amudhan et al., 2012</xref>).</p>
<p>Beberapa penyelidikan bioaktivitas menggunakan pelarut air dan etanol, biji pinang telah menunjukkan aktivitas antimikroba terhadap mikroba patogen. Menurut Peng et al (<xref ref-type="bibr" rid="redalyc_5043270002_ref20">2015</xref>) senyawa ekstrak pinang memiliki banyak bioaktivitas, termasuk sebagai antijamur. Penelitian Joseph &amp; Singh (<xref ref-type="bibr" rid="redalyc_5043270002_ref13">2008</xref>) menyatakan ekstrak pinang menggunakan pelarut air terhadap <italic>C. albicans</italic> dengan konsentrasi 50 μl memiliki rerata daya hambat 18 mm, dalam studi yang sama ekstrak kloroform melalui metode sokhlet dengan taraf konsentrasi yang sama memiliki rerata daya hambat sebesar 23 mm. Selanjutnya Penyelidikan lain terhadap ekstrak buah pinang menunjukkan, ekstrak biji pinang memiliki aktivitas penghambatan terhadap <italic>C. albicans</italic> (<xref ref-type="bibr" rid="redalyc_5043270002_ref5">Anthikat et al., 2014</xref>; <xref ref-type="bibr" rid="redalyc_5043270002_ref18">Pahadia et al., 2013</xref>).</p>
<p>Senyawa flanovoid dalam biji pinang diantaranya adalah quercetin (<xref ref-type="bibr" rid="redalyc_5043270002_ref30">Yang et al., 2012</xref>). Quercetin memiliki aktivitas anti jamur yang bermanfaat dalam pengelolaan klinis vaginitis Candida yang disebabkan oleh biofilm <italic>C. albicans</italic> dan merupakan agen sinergis yang menjanjikan bersama dengan flukonazol (<xref ref-type="bibr" rid="redalyc_5043270002_ref10">Gao et al., 2016</xref>).</p>
<p>Salah satu senyawa penting dari kelompok tanin dalam biji pinang adalah senyawa katekin dan epikatekin (<xref ref-type="bibr" rid="redalyc_5043270002_ref2">Amudhan et al., 2012</xref>; <xref ref-type="bibr" rid="redalyc_5043270002_ref3">Ansari et al., 2021</xref>). Senyawa ini banyak ditemukan pada tanaman teh dan telah dilakukan kajian efek farmakologis antimikrobanya, termasuk kajian sebagai kandidat anti jamur. Riset Sitheeque et al (<xref ref-type="bibr" rid="redalyc_5043270002_ref25">2009</xref>) tentang katekin bersama theaflavin pada konsentrasi 6,25 ml memiliki daya hambat terhadap jamur <italic>C. albicans.</italic>
</p>
<p>Selain itu asam lemak yang terkandung dalam biji pinang, seperti <italic>gallid acid, lauric acid, decanoid acid, myristic acid</italic> dan <italic>tetradeconoid acid</italic> (<xref ref-type="bibr" rid="redalyc_5043270002_ref2">Amudhan et al., 2012</xref>; <xref ref-type="bibr" rid="redalyc_5043270002_ref3">Ansari et al., 2021</xref>) sangat berpotensi menjadi senyawa yang dapat menghambat pertumbuhan jamur <italic>C. albicans. Gallic acid</italic> salah satu senyawa yang banyak ditemukan pada berbagai tumbuhan. Efikasi asam lemak <italic>gallic acid</italic> tanaman <italic>Punica granatum</italic> secara in vitro dapat menghambat pertumbuhan <italic>C. albicans</italic>. Penelitian lain menunjukkan <italic>gallic acid</italic> yang diisolasi dari <italic>Buchenavia tomentosa</italic> menunjukan penghambatan pada plantonik jamur patogen <italic>C. albicans</italic> (<xref ref-type="bibr" rid="redalyc_5043270002_ref27">Teodoro et al., 2015B</xref>). Selanjutnya gallic acid sangat potensial untuk dikembangkan sebagai senyawa anti jamur untuk kepentingan klinis (<xref ref-type="bibr" rid="redalyc_5043270002_ref16">Li et al., 2017</xref>).</p>
<p>Selain itu <italic>lauric acid</italic> dan <italic>decanoid acid</italic> memiliki aktivitas antijamur terhadap <italic>C. albicans </italic>(<xref ref-type="bibr" rid="redalyc_5043270002_ref7">Bergsson et al., 2001</xref>; <xref ref-type="bibr" rid="redalyc_5043270002_ref14">Kabara et al., 1972</xref>; <xref ref-type="bibr" rid="redalyc_5043270002_ref17">Murzyn et al., 2010</xref>)<italic>. Myristic acid</italic> dan <italic>tetradeconoid acid</italic> juga dapat menghambat jamur <italic>C. albicans</italic> (<xref ref-type="bibr" rid="redalyc_5043270002_ref14">Kabara et al., 1972</xref>). Mekanisme kerja daya hambat asam lemak terhadap jamur diantaranya merusak membran sel, menghambat β-oksidasi, sintesis triacylglycerol dan sphingolipid dan kemungkinan menghambat aktivitas enzim topoimerase (<xref ref-type="bibr" rid="redalyc_5043270002_ref22">Pohl et al., 2011</xref>).</p>

<title>
<bold>KESIMPULAN DAN SARAN</bold>
</title>
<p>Ekstrak etanol biji pinang memiliki kemampuan menghambat pertumbuhan jamur <italic>C.albicans</italic>. Ragam senyawa metabolit sekunder yang terkandung didalamnya seperti flavonoid, terpenoid, tanin, dan beberapa asam lemak diduga memiliki aktivitas antijamur. Biji pinang dapat dijadikan sebagai alternatif atau kandidat pengembangan obat antijamur.</p>

<title>
<bold>Kekurangan Penelitian</bold>
</title>
<p>Penelitian ini memiliki kekurangan sampel penelitian yang hanya menggunakan 3 ulangan, menggunakan aquades sebagai pelarut. Data penelitian tidak terdistribusi normal sehingga uji statistiknya menggunakan uji non parametrik.</p>
